Description
SS-31, also known as elamipretide, is a Szeto-Schiller peptide with the sequence D-Arg-Dmt-Lys-Phe-NH2, where Dmt is 2′,6′-dimethyltyrosine. Its alternating aromatic and cationic residues give it a net positive charge at physiological pH while still allowing it to cross cell membranes, and it accumulates at the inner mitochondrial membrane.
Mechanistic studies have shown that SS-31 associates with cardiolipin, the phospholipid that supports cristae architecture and the organisation of electron-transport-chain complexes. Preclinical work has used it to examine mitochondrial reactive oxygen species, the peroxidase activity of cytochrome c and cristae structure in cell models, and in rodent models of cardiac, renal and neuronal mitochondrial dysfunction. Its small size and defined sequence also make it a convenient reference compound when comparing other mitochondria-targeted peptides.
